Cancer is the alteration of DNA signaling and out of control growth, altered DNA caused by tobacco usage, alcohol abuse, radiation and radon exposure, ultra violent rays from sunshine and exposure to toxins like aflatoxin and other carcinogens. Along with other proteins, essentially monitors the health of cells, cellular DNA and when irregularity is sensed, p53 aid in deciding whether to repair or begins apoptosis. As a transcription factor, p53 initiates the transcription of a group of targeted genes. One of the most important of these genes is p21. The transcription of p21, thereby, ultimately prevents all proliferation. Because of this, the cells are given the opportunity repair damages; and if this damage is irrepairable and there is significant alterations, p53 will trigger apoptosis and prevent the damage cell from responding. It is also important to note, however, that a cell lacking p53 does not automatically lead to cancerous cells; though it certainly increases the likelihood for a cell to be cancerous cancer.
Thereby, a dysfunctional p53 would lead to the accumulation and such mutations and damages. Mutations can occur in regulatory regions in the p53 gene that can lead to a number of consequences, depending on the extent and location of alterations. These areas regulate gene transcription in a region called, “the promoter area” which can result in the lack of p53 as we start figuring out what makes cancer tick.
Mutations can also occur in the protein coding region of the gene which would then alter the activity of the protein. There would be a decrease in the role p53 as a transcription factor. Hence, a mutation in the p53 gene can lead to LI – Fraumeni Syndrome; which occurs when an off – spring receives a defective copy of the p53 gene. This often leads to various types of cancers, including breast and thyroid cancers at very young ages.
Studies have shown that p53 drug therapy is one very effective treatment for cancer, which p53 is administered in the form of the drug Advexin. Advexin, injects p53 protein, in high concentrations into cancer tissue that cancer cells are selectively destroyed and the surrounding cells remains unharmed.
